The animal-free, recombinant TrypsiNNex® enzyme has a sequence identical to porcine trypsin, making it a direct substitute when switching to animal-free manufacturing. Produced in E. coli in an inactive form, the enzyme remains inactivated throughout upstream and downstream processing. This approach minimises auto-degradation before final stabilisation and packaging, preserving a high proportion of β-trypsin.
By nature, trypsin undergoes autolysis in a series of steps: β-trypsin is gradually converted into α-trypsin via proteolytic cleavage, and intermediate forms such as Ψ-trypsin may appear. These intermediates have less specific activity, which is undesirable in controlled bioprocesses.
A high β-trypsin level is therefore a key quality marker, ensuring reproducible activity and high specificity.
TrypsiNNex® typically exceeds the minimum β-trypsin specification of 70%, delivering reliable activity and a consistent proteolytic profile — an essential factor when processing precursor proteins like pro-insulin.

- Incoming testing of TrypsiNNex® (e.g. identity, activity)
- Detection of residual raw material in the finished product (LOQ for HPLC-based purity measurement)

Although there are no specific regulatory guidelines at this time to specify allowable amounts of residual Trypsin or Collagenase, these are still considered process residuals and potential product contaminants, and thus should be assessed as part of an overall risk mitigation strategy.
