Insulin is used to optimise cell culture media systems for a range of cell lines, including CHO-S, CHO-K, HEK-293, and Sf9. As a result, it is used to improve the upstream manufacturing of monoclonal antibodies, virus vaccines, and viral vectors.
When applied to a fed-batch culture of CHO-K1 cells for monoclonal antibody (mAb) production, insulin can be used to increase yields through the basal medium and through the feeding medium. In a two-part experiment, researchers found that 2 mg/L insulin in the basal medium increased cell growth by 17% and antibody production by 51%. Meanwhile, supplementing the feeding medium with 2 mg/L insulin improved antibody production by 49%.
Data also supports the use of recombinant insulin in gene therapy manufacturing, where it can help manufacturers scale up viral vector production. Supplementing HEK293 cells with Insulin Human AF resulted in a 68% increase in titres of AAV2, AAV5, and AAV8. In addition, when tested in a HEK-293 cell line for lentivirus production, a 120% titre increase followed.
Recombinant insulin can also help the viral vaccine industry meet high demand. In HEK-293 cells, Insulin Human AF supplementation produced a four-fold increase in cell growth and a 60% increase in virus production.
